    I've a 3d printer that I've recently built using V slot.

It's been continually printing for around 30 hours, and I noticed some white film(like spider web) kind of substance in all Y axis v wheels but couldn't find anything on X axis

My X & Y axes Universal gantry plate running on 20x40 V slot using V wheels

For X axis gantry plate is placed vertically and for Y gantry plates are placed horizontally

Since I could only find this in Y axis, Is V wheel + gantry plate not designed build in horizontal manner as seen in picture?

3D printers tend to create "spider webs". Could be attracted to that area by static or perhaps air flow.

One thought would be that you have too much pre-load on the wheels. Where did you source your wheels from?

I've not sourced it from Openbuilds, since I live in India. I bought it from an online store 3DPrintronics

My gantry assembly has Universal gantry plate + Spacer block+ 6.35 mm spacer(2X)+6.35 mm eccentric(2x)

When I was assembling, even when the eccentric nuts positioned give maximum offset, the wheels were little tight on V slot rail. It could be due to that.

Pre-load: If I understand that correctly, In the case of V-slor + V wheels, how wheels are positioned to reduce play by adjusting eccentric, right?

But it's not present on X and present on both Y axes

I think more photos are called for here.
Are you sure that is V Slot?
If not, the sharp edge would be cutting into the plastic.

    Correct.

I'm guessing the problem is probably a combination of the pre-load and low quality wheels. Good quality acetal wheels shouldn't be doing this. They will develop temporary flat spots with to much pre-load but I've never heard of them shredding. You might see if you can source some better quality wheels if the problem continues.
